Segregate embryonic lethals (n499 homozygotes) and WT. Remove WT from plate to prevent them from taking over. This strain is extremely difficult and time consuming to maintain. Gives very few Uncs. Heterozygotes tend to stack up in parallel clumps. Populations can be enriched by transferring these clumps to new plates and allowing wild-type to crawl out into bacterial lawn, and then picking away wild-types or transferring the clump of Hets to another plate.
