Modular linker::3xGFP11::3xFLAG::linker tag inserted at the C-terminus of the endogenous col-61 locus by CRISPR. Allele obtained using Cas9 RNP. Cassette design allows for re-editing of locus with common crRNAs/sgRNAs. Derived by sequential edits of parental strain NM5546: wrdSi133 was an insertion into jsSi1726, and subsequent excision left wrdSi142 [loxP::eft-3p::ssGFP1-10::ubl-1 3'UTR::FRT3] inserted at the former jsSi1726 site.
