Descriptionrde-4(ne301); qtEx136[rgef-1p(F25B3.3)∷unc-22sense; rgef-1p∷unc-22antisense; rgef-1p∷DsRed line 8]; jamEx3[rgef-1p::rde-4(+)::rde-4 3’UTR & rgef-1p::gfp::unc-54 3’UTR] animals were crossed with WM49 (rde-4(ne301) III) animals. rde-4(ne301); jamEx3[rgef-1p::rde-4(+)::rde-4 3’UTR & rgef-1p::gfp::unc-54 3’UTR] animals were isolated. This strain is expected to drive the expression of RDE-4 under the control of the neuronal promoter rgef-1p from an extrachromosomal array in an rde-4(ne301) background, which could result in wider expression of RDE-4 because of the unknown complexity of extrachromosomal arrays. GFP+ animals from this strain show silencing of muscle, intestine, and hypodermal genes by dsRNA. Pick GFP+ animals to maintain the extrachromosomal array.
