pat-3(st564) III; kqEx73. |
kqEx73 [pat-3(sp) + rab-3::RFP + cki-1::GFP]. Pick RFP+ to maintain. kqEx73 carries a form of pat-3 gene with splicing defects; rescues pat-3 null allele. pat-3(sp) is a frameshift mutation in the splice acceptor region (ag to aa) that abolishes conserved interaction domains such as the NPxY motifs and creates a splice variant with an extra 19 amino acids. The pat-3(sp) animals not only produce mutant pat-3, but also express the regular splice form due to utilization of an unusual splice acceptor. Abnormal Distal Tip Cell migration and pat-3 gene splicing (intron 7) defects. Reference: Kihira S, et al. PLoS One. 2012;7(8):e42425. |

pat-3(st564) III; mwEx32. |
mwEx32 [pat-3(YYFF) + sur-5::GFP]. Pick GFP+ to maintain. mwEx32 carries a mutant form of pat-3 gene with tyrosine (Y) to phenylalanine (F) mutations; rescues pat-3 null allele. Abnormal DTC migration. Reference: Lee M, et al. J Biol Chem. 2001 Sep 28;276(39):36404-10. |

pat-3(st564) III; mwEx443. |
mwEx443 [pat-3(+) + sur-5::GFP]. Pick GFP+ to maintain. mwEx443 contains wild type contains pat-3 genomic DNA, rescues pat-3 null allele. Reference: Lee M, et al. J Biol Chem. 2001 Sep 28;276(39):36404-10. |

pat-3(st564)/qC1 [dpy-19(e1259) glp-1(q339)] III. |
Heterozygotes are WT and segregate WT, DpySteriles and PATs. st564 is a recessive lethal which causes a "severe" PAT phenotype. Strain is well balanced. |

pat-3(st564)/qC1 [dpy-19(e1259) glp-1(q339)] III; mir-83(n4638) IV; mir-34(gk437) X. |
Heterozygotes are superficially WT (with DTC migration defects), and segregate superficially WT (with DTC migration defects), sterile ts-Dpy qC1 homozygotes, and st564 homozygotes (PAT lethal). Pick WT and check for correct segregation of progeny to maintain. Reference: Burke SL, Hammell M, Ambros V. Genetics. 2015 Jun 15. |
